Crestor is usually prescribed to lower the risk of heart attack and stroke due to high cholesterol levels in the blood. It is indicated in the following cardiac disorders:
High blood cholesterol levels (Hypercholesterolemia)
Hereditary or genetic high blood cholesterol, fats levels (mixed Hyperlipidemia)
Homozygous and Heterozygous familial Hypercholesterolemia (HeFH)

Dose, Administration and Dosage forms:
Crestor is available in tablet formulation in 5mg, 10mg, 20mg and 40mg dosages. Crestor may be taken with or without food. The usual dosage may vary from 5mg to 40mg once per day depending on the severity of the patient's condition. The higher dose 40mg daily is used in those patients not reaching LDL-C goal with lower doses. In pediatric patients with HeFH, the common dose is 5mg to 20mg daily.
Mechanism of action:
Crestor is reported to be an HMG-Co-A reductase inhibitor. Crestor acts by selectively and competitively inhibiting the HMG-Co-A reductase which is a rate limiting enzyme. This enzyme is responsible for the conversion of 3-hydroxy-3-methylglutaryl coenzyme A into mevalonate which is a precursor of cholesterol (LDL and VLDL) production in the human body. Once this enzyme is inhibited by Crestor, bad cholesterol and triglyceride production decreases, resulting in reduced risk of cardiac diseases related to high cholesterol levels. Crestor also helps increase hepatic LDL receptors to enhance uptake and catabolism of LDL. This also inhibits VLDL synthesis resulting in suppressed VLDL and LDL. Crestor also helps improve good cholesterol (HDL) levels.
Side-effects and safety profile:
Common side-effects associated with Crestor usage include muscle pain, tenderness, weakness, and fever. Crestor may also result in some rare but severe liver problems like yellowing of the skin and eyes, and darkening of the urine. It may also cause some gastro-intestinal issues such as severe abdominal pain, persistent nausea, and vomiting. Some patients may experience allergic reactions to Crestor such as itching; rash; facial, tongue or throat swelling; and trouble breathing.
If you are allergic to Crestor, you should not take this medication.
If you have had prior trouble with liver function, then you must take Crestor carefully, keeping in continuous touch with your physician in order to avoid any unwanted reactions.
Inform your physician about other medications you are taking before starting Crestor so that any drug interaction can be avoided. Common drugs that are contraindicated with Crestor are Ciclosporin, Bezafibrate, and Gemfibrozil.
Avoid driving and operating machinery, as Crestor may cause dizziness.
Patients on higher Crestor dosing (40mg dose) must be monitored for proper kidney function.
If you have a history of kidney, liver, or muscle conditions, you must closely monitor any side-effects that Crestor may present.
You should not take Crestor if you currently have liver, kidney or muscular disease.
Avoid taking Crestor if you are pregnant or breastfeeding.
Crestor is primarily prescribed to adults who need help regulating their cholesterol. This drug is used to lower LDL cholesterol and raise HDL cholesterol. The liver produces much of the cholesterol that is found in the body. The main ingredient of Crestor, rosuvastatin calcium, works by inhibiting a specific enzyme in the liver that is crucial to the production of cholesterol. By adjusting cholesterol levels, Crestor helps to inhibit the buildup of plaque in the arteries, a disorder known as atherosclerosis. Crestor is most successful when used in conjunction with a healthy diet and regular exercise.
